Do health insurance companies like Blue Shield of California require blood test for coverage?

1. They could require it. Any insurance company whose "numbers" are based on the life (and health) of a person, might require an extensive medical study of that person.
2. Most health insurance companies don't require a blood test unless there is a questionable health condition or a past history of alcohol or drug abuse. I've never heard of any company requiring a test for any other reason.
